 BACKGROUND: There has been an increase in the incidence of occupational asthma along with better understanding of its pathophysiologic mechanisms and etiologic factors. There are no reports of patients with asthma and rhinitis to Passiflora alata (passion flower) and Rhamnus purshiana (cascara sagrada). METHODS: We describe two substances of plant origin as causal agents of occupational allergic respiratory diseases in a patient who worked in a pharmacy devoted to the manual preparation of products. RESULTS: Skin testing and Western blot confirmed the sensitization of the patient to these plant extracts in vivo and in vitro, respectively. Bronchial challenge confirmed the cause-effect relationship between the allergen exposure and the diseases. CONCLUSIONS: We conclude that Passiflora and cascara sagrada are two new etiologic agents of IgE-mediated occupational asthma and rhinitis. The present study also serves to alert physicians to the risks associated with work in pharmacies devoted to manual preparation of plant extracts, emphasizing the importance of the use of protective measures in these environments.

 Tissue cultures initiated from the cambial zone in branches of RHAMNUS FRANGULA L. and RHAMNUS PURSHIANA DC were grown on modified Murashige and Skoog medium with kinetin and 2,4-D as growth factors. To study the production of the genuine anthraquinone derivatives, the extraction of the fresh calli was performed in such a way that oxidation was prevented. In the fresh callus of RHAMNUS FRANGULA the glycosides of chrysophanoldianthrone and chrysophanolphys-ciondianthrone predominantly occurred, whereas in the freeze dried bark of RHAMNUS FRANGULA the glycosides of emodin and emodinanthrone were the main components. Aloe-emodin, which does not occur in the intact bark material of RHAMNUS FRANGULA, was proved to be present in the callus culture. In the fresh callus of RHAMNUS PURSHIANA the glycosides of chrysophanolanthrone and physcionanthrone dominated, while in the freeze dried bark of RHAMNUS PURSHIANA the glycosides of emodin, emodinanthrone and aloe-emodin were predominantly present. The total yields of the glycosidic bound aglycones produced in both cultures were approximately one third of the yields produced in the bark of the two intact species from which the cultures originated.

  Colon cleansing regimens. A clinical study in 1200 patients.:

 The purgative effect of bisacodyl, anthraquinone glycosides ( Cascara sagrada ), and sodium picosulfate, alone or in combination with a saline purge and a tap water enema, was studied in 1200 patients. The cleansing effect was scored with regard to retained fecal residue evident on double-contrast studies of the colon. The combination of a contact laxative and a saline purge produced good cleansing effect in 52%-80% of the patients. With an additional tap water enema given 1 hour before the colon examination, however, 96% of the colons were clean. The taste and the effects of the cleansing systems were tolerated favorably by more than 90% of the patients. However, 17% reported restriction in work capacity on the day of bowel cleansing.

  Simultaneous determination of resveratrol, emodin, chrysophanol, physcion, in root of Polygonam cuspidatum and its extract by HPLC.:

 Objective: To establish a HPLC method for simultaneous determination of resveratrol, emodin, chrysophanol, physcion in root of Polygonum cuspidatum and its extract. Method: The separation was performed in a Hypersil ODS C18 column with a mobile phase of Acetonitrile-1%HAc. The flow rate was 1.0 mL x min(-1) and detection wavelength was at 287 nm. RESULT: The mean recovery and RSD of resveratrol, emodin, chrysophanol, physcion was 101.8%, RSD 1.7%; 96.2%, RSD 1.4%; 99.7%, RSD 1.0%; 98.60%, RSD 0.97% respectively. CONCLUSION: The method is accurate, simple and reliable. It can be used for quality control of P. cuspidatum and its extract.

  Preparative isolation and purification of five compounds from the Chinese medicinal herb Polygonum cuspidatum Sieb. et Zucc by high-speed counter-current chromatography.:

 High-speed counter-current chromatography (HSCCC) was applied to the separation and purification of five compounds from the Chinese medicinal herb Polygonum cuspidatum Sieb. et Zucc. The crude extracts from P. cuspidatum Sieb. et Zucc were treated with light petroleum-ethyl acetate-methanol-water (2:5:4:6, v/v). Sample 1 was obtained from the lower phase and sample 2 from the upper phase. The sample 1 was separated with light petroleum-ethyl acetate-water (1:5:5, v/v) and yielded 19.3mg of piceid, 17.6 mg of anthraglycoside B from 200mg of sample 1. The sample 2 was separated with light petroleum-ethyl acetate-methanol-water (3:5:4:6, v/v) and light petroleum-ethyl acetate-methanol-water (3:5:7:3, v/v) in a gradient elution and yielded 18.5mg of resveratrol, 35.3mg of emodin and 8.2mg of physcion from 220 mg of sample 2. The purity of each compound is over 95% as determined by HPLC. The chemical structures of these components were identified by (1)H NMR and (13)C NMR.
